    American Cancer Society Research Scholar Grant       $720k total over 4 years ($150k/yr dir)
    Improved MR spectroscopic imaging for glioma treatment planning
    We propose a novel algorithm to directly estimate the metabolite concentrations from MRSI data, thereby improving the robustness and sensitivity to J-coupled metabolites. We will test the utility of the J-coupled metabolites in determining the microscopic spread of brain cancer, with the focus of using the information for radiation treatment planning.

    Ocala Royal Dames for Cancer Research       $50,000 total over 2 years
    Phase II surveillance imaging trial for distant metastases in Stage III & IV breast cancer patients
    Goal to perform surveillance CT imaging of the chest and abdomen to identify early stage metastases to the lung, liver and thoracic boney anatomy. Randomized treatment trial involved using surveillance imaging of 12 subjects with recommendation for localized, targeted, ablative therapy of any identified lesions. This is a 2-arm study. Because the number of subjects is small, the endpoint of demonstrating improved overall survival may not be achieved, but we expect 3 subjects to experience prolonged overall survival compared to controls. Important information to be gathered include the number of and size of metastases at first incidence, the time to first incidence, and number of patients presenting with new lesions within the 2-year time period of this award.

    Computer-aided detection of polyps in CT Colonography
    A novel three-dimensional (3-D) computer-aided detection system developed in-house has been developed and demonstrated to be particularly efficacious for lung and brain metastases. Our hypothesis is that with appropriate customization this system will achieve detection outcomes with CTC data that are comparable to those currently experienced in optical colonoscopy.

    American Heart Association Western States Affiliate Postdoctoral Fellowship
    Myocardial Architecture and Regional Function using MRI
    The first aim of this project was to measure the principal axes of water diffusion in excised strips of myocardial tissue and correlate that with histological measurements of local fiber orientations. The second aim was to perform diffusion MRI experiments in a passively deforming intact canine heart preparation to assess changes in fiber orientation during deformation. The third aim was to integrate fiber architecture within a three-dimensional finite element model of a passively deforming left ventricle. The PI was involved in all aspects of this project.

    American Cancer Society Research Scholar Grants for Beginning Investigators     $391,000
    Gated External-beam Radiotherapy of Isolated Tumors in the Lung and Liver
    This project uses magnetic resonance imaging of liver and lung tumors in patients to establish both the range of motion of these tumors during the respiratory cycle, and determine the reproducibility of tumor position over multiple end-expiration breath-holds. This information will be used to develop analysis tools and motion phantoms to test feasibility of performing gated radiotherapy of internal tumors, obtain physiologic measurements of respiratory-derived tumor motion, and establish the performance characteristics required of a real-time gating system.

    Quantifying Human Lung Tissue Dose Response Relationship
    This project proposes to develop a novel tool for quantifying the radiation dose-response relationship for normal lung tissue in human subjects treated for metastatic lung lesions using conformal, high-dose stereotactic radiotherapy. This research involves the development of image processing methods for extracting radiographic changes in post-treatment subjects and correlation with radiation dose profiles retrieved from treatment planning systems. An animal study of the histological basis for the observed radiographic changes is also proposed to related radiation levels to subsequent cell survival statistics.

    Cancer Treatment Research Foundation     $240,000 direct/yr
    Predicting Brain Tumor Recurrence from MR Spectroscopy and Diffusion Tensor Imaging
    Stereotactic radiotherapy (SRT) treatment plans of aggressive brain tumors typically incorporate a uniform 2 cm margin to account for unobserved, microscopic dispersal of cancer cells, yet recurrent tumors often occur. Our hypothesis is that brain cancer cells spread preferentially via a diffusion-like process that can be predicted by magnetic resonance (MR) diffusion-tensor imaging (DTI) and computational models of cell migration. We will also test whether MRS can be used to identify existing microinfiltration of cancer. The goal is to apply both MRS amd DTI to develop appropriate non-symmetric margins for radiation treatment of malignant brain tumors. The aims of this project are to gather prospective MRS/DTI data and patient follow-up MRI's to access the predictive power of MRS/DTI for identifying the locations of subsequent tumor recurrence. The second aim is to determine the efficacy of appropriately altering SRT treatment margins in the clinic.

    MR spectroscopic imaging of brain cancer progression
    The main objective of this proposal is to further develop this technology, with the focus on brain cancer progression and treatment planning. The specific aims are (a) to develop a joint reconstruction and quantification algorithm, thus further improving robustness and reducing artifacts (b) to optimize the multi-parametric sequence to enhance the sensitivity to metabolites crucial in brain cancer (c) to study the brain cancer progression and effectiveness of therapy.     Monitoring of Tumor Recurrence: DTI vs Histology
    Our hypothesis is that brain cancer cells spread preferentially via a diffusion-like process that can be predicted by magnetic resonance (MR) diffusion-tensor imaging (DTI) and computational models of cell migration. This pilot proposal will use a native rat infiltrative glioma cell line to validate the role of DTI in predicting tumor spread in the in-vivo rat brain and to optimize the computational model of cell migration for application to human subjects. RCBI provides 10 scan sessions and RF coil development.

    Our hypothesis is that brain cancer cells spread preferentially via a diffusion-like process that can be predicted by magnetic resonance (MR) diffusion-tensor imaging (DTI) and computational models of cell migration. This pilot proposal will fund the animal procurement, care and engraftment of a native rat infiltrative glioma cell line to optimize our computational model of cell migration.
